The inhibition activity of selected ß-carboline alkaloids on enzymes of acetylcholinesterase and butyrylcholinesterase

This thesis deals with testing of inhibition activity ß-carboline alkaloids on activity of enzymes acetylcholinesterase (ACHE) and butyrylcholinesterase (BUCHE) using test "Fast Blue B salt" at TLC desk and Ellman's test using spectrophotometer. It was also investigated how dimethylsulfoxide used as a solvent in combination with water affects activity of enzymes and alkaloids. Influence of membranes on alaptide permeation from hydrogels

The paper evaluates the liberation of alaptide from gels through various types of permeable membranes. The gels were prepared on the basis of three different polymers (3% chitosan; 2.5% hydroxypropyl cellulose; 3% hydroxyethyl cellulose) in different concentrations with additions of humectants (5 %; 15% propylene glycol and 10% glycerol) and the preserving agent, 0.3% Sepicide HBR with 1% alaptide, and finally without the drug. The paper examines the effects of changes in medicinal drug policy valid since 1 January 2008 by comparing the dispensation to patients in a pharmacy supplying a hospital in the years 2007 and 2008. The examination was performed using financial representation (sale price without VAT), the number of original packages and classification according to ATC groups. The data were obtained from the information system of the pharmacy. Since 1 January 2008, the health service reform established the duty to pay a regulatory fee for the dispensation of one item on the prescription which is at least partially covered by the public health insurance and it also established a protective annual limit of the sum of a patient's financial participation in the provided health care. The study aimed to determine the effects of these measures. The complete medication of 100 patients for the year 2008 was examined. Inter-individual variability in drug response is a major clinical problem. Much of the variability has been observed in drug metabolism, particularly in the enzymes of cytochrome P450. Genetic polymorphism in these enzymes may influence a patient's response to commonly prescribed drugs. The first part of this review describes the enzymatic system of cytochrome P450 and further focuses on the influence of genetic polymorphism of cytochrome P450 1A2 on drug effect.
