Soft Gelatin Capsules, Their Manufacture and Evaluation

J. Kolčák, M. Stuchlík, M. Rabišková

Čes. slov. farm. 2003, 52(1):4-11

Soft gelatin capsules are a dosage form suitable particularly for administration of sensitive drugsposing technological problems. They mask an unpleasant taste or smell of the drug and the colourcombinations increase the safety of therapy by excluding interchangings. Their manufacture isundemanding, it depends on the quality of gelatin and the gelatin mass for the shell of the capsules,on the properties of the fill, on mutual interactions between the fill and the shell, and on a specialknow-how, which is an unconditional precondition of successful manufacture of this dosage formThe paper is a review characterizing soft gelatin capsules, processes of...

Chalcones (1,3-Diarylpropen-1-ones) and Their Analogues as Potential TherapeuticAgents of Diseases of the Cardiovascular System

V. Opletalová, L. Jahodář, D. Jun, L. Opletal

Čes. slov. farm. 2003, 52(1):12-19

At the beginning of a new millennium, several important questions have come to the fore connectedwith the utilization of natural resources for the investigation of new medicaments (necessity ofcooperation of both highly developed and developing countries in the protection of natural resources, necessity to justly evaluate and reward the contributions of the individual cultures in the case thata knowledge of traditional medicine was employed in the development of a new medicament, necessity to achieve the maximal chemical variety of real and virtual libraries of substances fromwhich potential drugs are selected for screening tests). The research of chalcones...

Therapeutic and Toxicological Aspects of Biological Activity of Saint-John's-wort (Hypericum perforatum)

M. Kapusta, J. Dušek

Čes. slov. farm. 2003, 52(1):20-28

Saint-John's-wort (Hypericum perforatum L.) has been used in traditional medicine since timeimmemorial and it has its position also in the contemporary list of medicinal plants of pharmaceuticalimportance. Though it originally comes from Europe and Asia, it has been domesticated and, thanks to its properties, has been cultivated in many other parts of the world. The herbaceous plantcontains a wide spectrum of substances, out of which anthraglycosides, flavonoids, and floroglucinderivatives participate in principal pharmacological effects. In recent years this plant has becomegreatly popular as an effective alternative means for the treatment of weak...

Kinetics of Release of Scopolamine Bromide from Hydrogel by a Filtering Membrane

Z. Zatloukal

Čes. slov. farm. 2003, 52(1):29-32

The hydrated filtration membrane renders possible uniform hydration of hydroxypropylmethylcellulosehydrogel with subsequent uniform release of dissolved scopolamine bromide. Exponentialdependence of the diffusion coefficient on the graded medium dimension of the pores of the testingmembrane makes possible the estimation of the reactive fractal dimension DR by means of themodified exponent (3 - DR).With increasing concentration of scopolamine bromide in xerogel (14.7to 77.8 mg/cm3), DR decreases within a range of 2.814 to 2.704. A comparison with the structure ofa suitable fractal (Menger sponge) leads to a conclusion about suitability of the use of...

Dissolution of Nimodipine in Aqueous Solutions of Hydroxyethyl-β-cyclodextrin and theSurvey of Nimodipine Solubilization with Cyclodextrins

F. Kopecký, B. Kopecká, P. Kaclík

Čes. slov. farm. 2003, 52(1):33-38

The results of a comprehensive study of the dissolution of the calcium antagonist nimodipine inaqueous solutions of nine cyclodextrins are reported. The used cyclodextrins were native β-cyclodextrin(β-CD), its derivatives hydroxyethyl-β-CD (HE-β-CD), three hydroxypropyl-β-CD (HP-β-CD)with various degree of substitution and methyl-β-CD (M-β-CD), native α-cyclodextrin (α-CD),hydroxypropyl-α-CD (HP-α-CD) and hydroxypropyl-γ-CD (HP-γ-CD). The nimodipine dissolution was studied as a function of time (up to 14 days) and cyclodextrin concentration up to 0.07 mol/l,excepting the less...

Seasonal Variation in the Haemolytic Activity of the Capitula of Bellis perennis L.

T. Siatka, M. Kašparová

Čes. slov. farm. 2003, 52(1):39-41

The drug Bellidis flos is used in therapy as a diuretic and an expectorant agent. Its principalconstituents are saponins. The paper examined the haemolytic activity of the drug obtained by thecollection of the capitula of Bellis perennis L. in a onemonth interval (from March to October) in thecourse of three years. The determination of haemolytic activity was carried out following the methodof Pharmacopoea Bohemoslovaca IV. It has been found that the haemolytic activity of the drugchanges in dependence on the time of collection of capitula during the year; it is lowest in March, then it increases, reaching the maximum in summer months (June, July, August),...

New Groups of Potential Antituberculotics: 3-(4-Ethoxythiocarbonylphenyl)-2H-benzoxazin-2,4(3H)-dithiones and 3-(4-Ethoxythiocarbonylphenyl)-4-thioxy-2H-benzoxazin-2,4(3H)-ones

K. Waisser, P. Holý, O. Bureš, J. Kuneš, J. Kaustová

Čes. slov. farm. 2003, 52(1):42-47

In connection with the search for new antituberculotics, the present authors prepared a group ofderivatives of 3-(4-ethoxythiocarbonylphenyl)-2H-benzoxazin-2,4(3H)-dithione and 3-(4-ethoxythiocarbonylphenyl)-4-thioxy-2H-benzoxazin-2,4(3H)-one. The modifications of substituents were carriedout on the benzoxazine ring. The study was based on the present authors' previous assumptionthat the substitution of the oxo group for the thioxo group was connected with an increase inantimycobacterial activity. The prepared substances were evaluated by the activity against Mycobacteriumtuberculosis, Mycobacterium kansasii, and Mycobacterium avium. However, it was...
