Generics, Alternatives, Substitution, Interchange

J. Kolář, M. Chalabala

Čes. slov. farm. 1999, 48(2):59-64

Linking up with the topical activities of the International Pharmaceutical Federation (FIP) indefining the concepts generic alternative, generic substitution, therapeutic alternative, therapeuticsubstitution and therapeutic replacement and the proposed policy, the paper describes the state anddevelopment in the field of generics. The attention is devoted to the position of generics in selectedcountries, their definitions, labelling, pricing and generic substitution.

Human Hepatocyt: I. Model for the Study of Xenobiotic Metabolism and Toxicity

P. Kosina, Z. Dvořák, D. Walterová

Čes. slov. farm. 1999, 48(2):65-71

Isolated human hepatocytes have become one of the most attractive experimental approaches to thestudy of the specific metabolic functions of the human liver, interactions between liver cells andinfectious agents, and metabolism and pharmacotoxicity of drugs. Particularly the primary cellculture model provides an in vitro system for investigating specific mechanisms in a preciselycontrolled conditions. In the present paper the legislative and ethical problems concerning availa-bility of human liver samples, techniques developed for isolation, preservation and cultivation ofhepatocytes are discussed. In addition, a comparison of human and rat hepatocyte...

Density of Lyophilised Ophthalmic Lamellae2. Use of the Correlogram of Xerogel Density

Z. Zatloukal

Čes. slov. farm. 1999, 48(2):72-74

The paper examines the vacillation of the density of the foil lyophilised from hydrogel containing40 g/l of hydroxypropylmethylcellulose (HPMC 4000). Lamellae of a diameter of 0.55 cm were formedto estimate the density of cm 2 of xerogel. In comparison with traditional regressive methods, theuse of a two-dimensional correlogram is recommended. The most important finding is the achieve-ment of random vacillation of density in a distance of ca 2 cm in connection with stamping lamellaefrom the lyophilised foil. The findings make it possible to optimise the procedure of pouring hydrogelfor lyophilisation with a subsequent decrease in density vacillation...

A Study of Manufacturing Defects of Hard Gelatine Capsules ETALOCK

M. Rabišková, B. Štěrbová, P. Valášek

Čes. slov. farm. 1999, 48(2):75-76

The capsules ETALOCK meet the requirements for hard gelatine capsules. The share of defectiveproducts in the process of their forming and filling ranges within 0.7-1.5 %. If the filling machineseliminate also capsules without evident defects, it is due to the imperfect technical conditions of thefilling machines.

Evaluation of Preparations of Natural Medicine by Means of a Special FluorescentTechnique

K. Dubayová, L. Lešková, J. Kušnír

Čes. slov. farm. 1999, 48(2):77-79

The present paper aims to find a method of comprehensive characterisation and definition ofcomplex mixtures. For this purpose the technique of spectral fingerprint shows most advantages. It has been applied to the definition of very similar herbal antitussic preparations containing anextract from the plantain plant (Plantago lanceolata). The synchronous fluorescent spectrum (Dl =15 nm), in contrast to the classic emission spectrum, can resolve the individual preparations. Thecomplete characteristic of the preparation is, however, provided only by the method of a specialfluorescent fingerprint, which developed from a combination of the technique of recording...

The Relationship of the Structure, Physico-Chemical Properties and Local Anaesthetic Activity in a Group of 1-Methyl-2-piperidinoethylester of 2-, 3- and 4-Alkoxyphenylcarbamic Acids

M. Pokorná, J. Čižmárik, E. Sedlárová, E. Račanská

Čes. slov. farm. 1999, 48(2):80-86

Within the framework of a systematic study of phenylcarbamic acid derivatives with local anaes-thetic effects, a series of 1-methyl-2-piperidinoethylesters of 2-, 3- and 4-alkoxy(methoxy to decyloxy)phenylcarbamic acids and an unsubstituted derivative, i.e. 1-methyl-2-piperidinoethylester ofphenylcarbamic acid, were prepared. In the prepared series of compounds, the mutual relationshipof the surface local anaesthetic activity and some physical and physico-chemical parameters (g, pKa,pH of turbidity, log P, p, RMr, log k') was studied and quantified by the method of QSAR analysis. The steric constants L and B4, molar refraction (MR), valence indices...

Preparation, Aggregative Properties and Antimicrobial Activity of N-Alkyl-N-(2-hydro-xydodecyl)-N,N-dimethylammonium Bromides

D. Sládková, I. Csiba

Čes. slov. farm. 1999, 48(2):87-90

The paper describes the preparation of a series of six new organic ammonium salts of amphiphilic character, derived from amine alcohol, of the general structure: CH3(CH2)9CH(OH)CH2N + (CH3)2R Br -R = C6 - C12 A part of the synthesis of the above-mentioned compounds is also the preparation of 1-bromo-2-dodecanol. The aggregation properties of these compounds in aqueous solution, expressed by the valuesof the critical concentration of micelle formation (Table 2), and antimicrobial activity, expressed bythe minimum inhibitory concentration (Table 3), in dependence on the step-by-step lengthening of the alkyl chain while preserving the principal structure...

Theory and Practice of Pharmacopoeial Control of the Quality of Drugs and AuxiliarySubstancesV. Rapae oleum hydrogenatum - A Proposed Test

J. Šubert, J. Kačuríková

Čes. slov. farm. 1999, 48(2):91-93

The quality indices of hardened rape-seed oil for pharmaceutical purposes which is used asa component of ointments have been formulated and substantiated. The proposal is concerned withtesting from the physico-chemical aspects.

A Contribution to the Analytical Profile of N-isopropyl-3-[2-acetyl-4-(propoxycarbonyl-amino)phenoxy]-2-hydroxypropylammonium Chloride (BL 343 Ac)

M. Blešová, J. Csöllei, P. Novomeský, R. Opatřilová

Čes. slov. farm. 1999, 48(2):94-96

The paper presents some procedures of the analytical evaluation of a newly prepared substance withbeta-adrenolytic activity. The substance was characterised by its melting point, solubility; theexperimental partition coefficient, dissociation constant, surface activity were determined. Theprocedures for identification, purity evaluation and determination of the content of the activeprinciple ( 1 H-NMR, 13 C-NMR, IR, UV spectra, thin-layer chromatography, acidimetric determina-tion in a non-aqueous medium, extraction titration in a heterogeneous medium, extraction-photo-metric determination) were elaborated.
