Study of biocompatibility of peritoneal dialysis solutions measured as in vitro cells viability

Nataliia Hudz, Anna Filipska, Nataliia Stepaniuk, Nataliia Dmytrukha, Raisa Korytniuk, Piotr P. Wieczorek

Čes. slov. farm. 2019, 68(4):161-172 | DOI: 10.36290/csf.2019.019

This paper presents the comparable viability study results of the HepG2 and Vero cells in the presence of traditional peritoneal dialysis (PD) solutions determined by three methods (3-[4,5-dimethylthiazol]-2-yl-2,5-diphenyl tetrazolium bromide (MTT), neutral red (NR) and sulforhodamine B assays) with establishing different correlations between viability and quality indexes of the tested PD solutions. The obtained results confirmed cytotoxicity of the PD solutions even compared with an isotonic solution of sodium chloride. PD solutions action resulted in a similar reduction in the HepG2 and Vero cells. Moreover, this research found that metabolic cellular...

Nephroprotective effect of N-acetylglucosamine in rats with acute kidney injury

Sergii K. Shebeko, Igor A. Zupanets, Olha O. Tarasenko

Čes. slov. farm. 2019, 68(4):173-179 | DOI: 10.36290/csf.2019.020

The article presents the results of the study of the nephroprotective effect of N-acetylglucosamine (NAG) under the development of experimental acute kidney injury (AKI). The study was conducted on a model of acute glycerol nephrosis in rats. NAG was studied at a dose of 50 mg/kg at daily parenteral administration during
1 week compared to quercetin, which was administered intraperitoneally at a dose of 34 mg/kg. The efficiency of the drugs was assessed by the functional state of animals, the renal excretory function and the nitrogen metabolism indices. The NAG effect on rats with AKI caused a reduction of the mortality rate, an increase in...

New approach for detoxification of patients dependent on benzodiazepines and Z-drugs for reduction of psychogenic complications

Kateřina Kubová, Aleš Franc, Jakub Vysloužil, Jan Šaloun, David Vetchý

Čes. slov. farm. 2019, 68(4):139-147 | DOI: 10.36290/csf.2019.016

Benzodiazepines (BZDs) and Z-drugs are strongly addictive substances, acting on identical GABA receptors. Detoxification should be long-term and gradual, usually by tapering a long-acting BZD (diazepam) but no suitable commercial pharmaceutic product exists with the necessary low drug content. This review describes the specific pharmacological aspects and comparisons of individual BZDs in relation to their effects and addictiveness. The success of the treatment relates to the patient's comfort during this process. Patients are typically afraid of switching to a more suitable long-acting BZD (diazepam), and become stressed during the tapering and anxious...

Plant α-amylase inhibitors and their effect on the utilization of polysaccharides contained in the diet

Slavomír Kurhajec, Aleš Franc

Čes. slov. farm. 2019, 68(4):148-156 | DOI: 10.36290/csf.2019.017

Development of civilization diseases such as diabetes mellitus, metabolic syndrome or obesity, enforces the increasing effort to find new drugs, especially from natural sources. These include α-amylase inhibitors, which break down polysacharides into simple sugars in the body of a healthy person. As this cleavage affects the level of blood sugar, which is sought to be therapeutically influenced, there is a growing interest in these substances. This review maps the types of amylase inhibitors, including their natural resources.

Theory and practice of pharmacopoeial control of quality of drugs and excipients X. Number of parallel determinations, processing of results and their use in the assessment of the content of active substances and excipients in the European Pharmacopoeia (Ph. Eur.)

Jan Šubert, Jozef Kolář

Čes. slov. farm. 2019, 68(4):157-160 | DOI: 10.36290/csf.2019.018

In the European Pharmacopoeia (Ph. Eur.) there is no prescribed number of parallel determinations of the content of active substances and excipients. The authors suggest adding at least three determinations. The results of parallel determinations suggest to test for outliers before using test based on the ratio of the range of results (p = 0.95) and not to compare individual results with the tolerance limits given in Ph. Eur., but with their arithmetic mean. Furthermore, they propose to extend the Chapter 5.3. of the European Pharmacopoeia so as to be applicable not only to bioassays, but also to chemical, physicochemical and physical assays and tests,...
