The effect of Crataegus extract on heart mitochondrial function
D. Majienė, J. Bernatonienė, R. Masteiková, K. Dvořáčková
Čes. slov. farm. 2008, 57(6):239-243
This study was aimed to investigate in vitro the influence of different concentrations of Crataegus fruit extract (70 ng/ml - 13.9 μg/ml of phenolic compounds (PC)) on oxidative phosphorylation in isolated rat heart mitochondria. The mitochondrial respiratory rates were determined oxygraphically by means of Clark-type electrode system with different substrates: pyruvate + malate, succinate and palmitoyl-L-carnitine. Our results demonstrate that Crataegus extract at doses under to 278 ng/ml of PC had no effect on mitochondrial functions. At concentrations of 278 ng/ml - 6.95 μg/ml of PC, extract stimulated the State 2 respiration rate by 11-27%...
Consumption of hipolipidemics in the Czech Republic in 2000-2007
R. Bartas, J. Kolář
Čes. slov. farm. 2008, 57(6):244-248
Hypolipidemics are a group of pharmaceuticals with an expressive portion on pharmaceuticals costs. Analysis of their consumption has been an object of repeated studies in our republic as well abroad. The present data on hypolipidemic drug utilization in 2000-2007 were obtained from the database of the State Institute of Drug Control. Utilization was expressed in defined daily doses (DDD) according to the ATC/DDD system, costs in Czech crowns. Utilization was expressed in relative values - DDD per 1 000 inhabitants per day (DID). The utilization of fibrates in the period under study is unchanged. The utilization of statins increased from 9.4 DID (2000)...
Testing the effect of 2', 3, 4'-trihydroxychalcone in experimental diabetes mellitus: a pilot study
L. Bartošíková, J. Nečas, T. Bartošík, M. Pavlík
Čes. slov. farm. 2008, 57(6):249-253
The effect of 2', 3, 4'-trihydroxychalcone was monitored in the experimental conditions of alloxan-induced diabetes mellitus. The animals were divided by random selection into 2 groups (n = 7). The treated group was given chalcone in doses of 10 mg/kg in Avicel, the placebo group was given only a solution of Avicel. The last group was intact. Selected laboratory parameters (glucose, urea, cholesterol, antioxidative enzymes, total antioxidative capacity, malondialdehyde in serum; diuresis, total glucose and protein losses through urine) were determined. Kidney tissue and pancreas samples were taken for histopathological analysis. The results revealed...
Determination of metoprolol and its metabolite α-hydroxymetoprolol in serum by HPLC method with fluorescence detection

Čes. slov. farm. 2008, 57(6):254-259
High-performance liquid chromatography method has been developed with a view of its future use in the phenotyping of CYP2D6 enzyme by metoprolol as a probe drug. Metoprolol, α-hydroxymetoprolol and the internal standard nadolol were extracted from serum with dichloromethane alkalinized with 1 mol/l NaOH. Chromatographic separations were performed on the reversed-phase column SupercosilTM LC-18 (15 cm × 3 mm, 5 μm) with the mobile phase containing acetonitril : methanol : water : triethylamine (15 : 5 : 80 : 0.1, pH 3.0) at a flow rate of 0.7 ml/min. Fluorescence detection (FL) was made at 230 nm (excitation) and 300 nm (emission). The total...
Studies of local anesthetics Part 185: Thermodynamic parameters of heptacainium chloride in the solution of NaBr
F. Andriamainty, J. Čižmárik, I. Malík
Čes. slov. farm. 2008, 57(6):260-264
The critical micellar concentration (CMC) of the local anaesthetic agent heptacainium chloride in the solution of NaBr was determined by the spectrophotometric method in the UV region of the spectrum at the temperature range of t = 20-40 °C and pH ≈ 4.5-5.0. The dependence of CMC on the temperature T formed a U-shaped curve with the minimum at the temperature of t = 25 °C. The CMC dependence on the temperature T was fitted by the second degree polynomical function. The obtained parabolic equations were applied to the "phase separation model" and the following thermodynamic parameters could be calculated: standard Gibbs free energy (∆G°),...
Optimization of the extraction method for the determination of methadone and its metabolite EDDP in urine by gas chromatography
R. Horáková, J. Valentová, I. Pechová, F. Devínsky
Čes. slov. farm. 2008, 57(6):265-268
The present study aimed to develop a suitable extraction method for the determination of methadone and it's metabolite EDDP in human urine by gas chromatography with a nitrogen-phosphorus detector (NPD). The extraction method was optimized in an effort to receive the most satisfactory yield of the mentioned compounds from human urine. The highest yields were obtained by alkaline extraction (~ pH 9) with dietylether. Proadiphene (PA) was used as the internal standard. The most efficient yields were determined as follows: 91 ± 5 % for methadone (MTD) and 86 ± 5 % for its metabolite EDDP. The detection limit (LOD) of MTD was 8 ng/ml, for...
The effect of lipophilic carrier concentration on hydrophilic-lipophilic matrix systems characteristics
K. Dvořáčková, M. Rabišková, R. Masteiková, L. Okáčová
Čes. slov. farm. 2008, 57(6):269-274
The present experimental paper aimed to examine the effect of lipophilic carrier concentration on diclofenac sodium prolonged release from hydrophilic-lipophilic matrix tablets with respect to the optimization of its dissolution profile in vitro. The lipophilic carrier was cetyl alcohol contemporaneously acting as the melting binder in thermoplastic granulation, povidone was used as the gel-forming agent, sucrose or lactose were fillers. The obtained results indicated that cetyl alcohol concentration influenced both characteristics of the granulates and the tablets. The dissolution profile of sucrose samples was affected statistically significantly...

Possible effects on the liberation of alaptid from dermal semisolid preparations
V. Šimunková, Z. Vitková, M. Potúčková, E. Tichý
Čes. slov. farm. 2008, 57(6):275-277
The paper examines the effects of routinely used humectants of glycerol (GL) and propylene glycol (PG) on the liberation of alaptid (ALA) from 3% (m/m) chitosan (CHIT) hydrogels in comparison with the liberation of alaptid (ALA) from cream. The contents of GL and PG in the individual hydrogels were 5%, 10%, 15% (m/m). Alaptid is a regeneratively acting synthetic derivative of prolyl-leucyl-glycin amide, which is for the time being used in veterinary medicine. Its content in the hydrogels and cream under study was 1% (m/m). The experiments were performed in vitro in a tempered permeating apparatus. The amount of the released ALA was measured spectrophotometrically...
